Sofia Sachpekidou completed in the year 1981 her undergraduate studies in Byzantine and Modern Greek Philology, specializing in Modern Greek Philology, at the Aristotle University of Thessaloniki. Seeking further academic development, she pursued advanced studies at the Institute of Byzantine and Modern Greek Philology at the Freie Universitaet in Berlin, which was then West Berlin. During her time there, from 1985 to 1990, she not only engaged in her studies but also contributed as a research collaborator.

In 1991 Sofia Sachpekidou returned in Greece and was appointed to the Department of Philology in the Aristotle University. Her role at the department involved taking charge of the Library of Medieval and Modern Greek Studies. Since 2013 she works at the Library of Classical Philology and Ancient History, maintaining her position as the head librarian.

 She also participated in collaborative publications. The first titled “Byzantinische Bleisiegel in Berlin (West)” in 1986, and the second “Graecogermania: Griechischstudien deutscher Humanisten = ελληνικές σπουδές την εποχή του γερμανικού Ουμανισμού: die editionstaetigkeit der Griechen in der italienischen Renaissance (1469-1523)” in 1989.
